Dark Souls gave me several along the pathway.
Asylum demon... I didn't see the side-door to escape until I'd already died over and over for 20min trying to kill him with a broken sword. I knew then that this was going to be a slightly different and potentiall more intense game than Demon's Souls had been.
The graveyard of skellies... it has to be the "first level" after I make it to Firelink, right?
Gargoyles... I was running around like a chicken with my head cut off up there. I still feel awesome whenever I ring that first bell.
Capra Demon... This bastard made me paranoid to enter any fog gate for the rest of my first playthrough. I still get intimidated whenever I come up to that point when I need to get the Depths key. Even last time I took them on, the Capra Demon and his dogs Night-at-the-Roxbury'd me to death in that first left hand corner.
Looking back and seeing I was trapped in Darkroot by a fog wall and seeing several invaders start popping up... yeah I flipped my wig.
Getting to the Great Hollow and looking down...
Getting to Ash Lake and realizing I had to climb back up the Great Hollow.
After my first attempt at fighting the Super Giants before Orny/Smough... I was not prepared for a variation and I still get a little nervous fighting these guys especially if I'm in human form. Getting invaded while fighting them is basically death for me, lol.
If I thought about my first playthrough, I'm sure I could come up with more. Great game.

When I arrived in Blighttown and my drake sword was broke. I didn't put any points into strength or dex, other than the required wielding points. So I had nothing literally to help me out.
God, that place was a nightmare. ><
